Techniques Used

In vivo techniques

  • Two-photon laser scanning microscopy to assess Ca2+ - gradients in spinal dorsal horn neurons in vivo. -more-
  • Labeling of lamina I projection neurons by injection of the retrograde tracer DiI into the rat brain stem.
  • Extracellular single cell and field potential recording in spinal dorsal horn.
  • Animal models of neuropathic and inflammatory pain
  • Behavioral testing

In vitro techniques

    • Spinal cord slice preparation, with long dorsal root attached, from rats and transgenic mice
    • Whole-cell patch-clamp recordings under infrared differential interference contrast (IR-DIC) video microscopy to study intrinsic properties of nociceptive neurons and synaptic plasticity in superficial spinal dorsal horn
    • Perforated whole-cell patch-clamp measurements in spinal cord slice preparations
    • Microfluorometric measurement of [Ca2+]i with 2-Photon-Laserscanning microscopy and conventional video microscopy
    • Dorsal root ganglion (DRG) preparation
    • Electrophysiological recordings from DRG neurons with sharp electrodes
