Identification of alpha-centractin (P61163) by LC-MS/MS.
The peptides from the protein digest were eluted from a trapping column at a flow-rate of 250nl/min and separated using a gradient from 0.2% formic acid and 3% acetonitrile to 0.2% formic acid and 45% acetonitrile over 12 minutes. Eight peptide targets were chosen for fragmentation per MS-scan, the used settings allowed to perform 10 MS scans and 80 MS/MS-scans per minute on average. The raw data were interpreted by the Spectrum Mill software, providing results as shown. The listed peptides were identified with the indicated scores, %SPI indicates the percentage of the spectrum intensity which was interpreted according to the sequence. The identified peptides are highlighted in red in the amino-acid sequence of the total protein, yielding a coverage of 20%. Of the first five peptides listed, the MS/MS-spectra are shown. Note that almost complete y- and b-series were obtained, resulting in highly confident data interpretation. Peptide scores are mainly calculated from the sequence tag length.
