Data entry into the program is simple, just follow these steps. Go to the 'Data Entry' sheet either by clicking on DATA ENTRY on the title sheet of HN-NonLin or on the corresponding tab at the lower margin of your screen. After your first tests you may want to save a template (containing the information which will not change in future tests, e.g. drug concentrations) for further use. In many cases the data (e.g. raw data from plate readers) may simply be transfered by copy and paste.
Assign a name to to the experiment. This could be the name of the isolate / strain / clone you are testing or anything else that all data from this plate have in common.
Assign a label to each column. If you are testing one isolate with a number of different drugs on the same plate this would be the name of the drug, if you test different isolates on the same plate, enter the names of the isolates.
Enter the raw data from your plate reader or your parasite counts here. Always start with the control (usually the highest count) in the topmost row. Many scintillation counters or ELISA plate readers will allow you to export the results to a text or spreadsheet file. These may then be copy-pasted into the data input form.
Enter the drug concentrations. The control (concentration 0) must be in the topmost row. If you are using two-fold serial dilutions, all you have to do is to enter the highest drug concentration in the lowest row, all other concentrations will automatically be calculated. You may change the dilution factor in the 'Options' sheet.
Once all your data are entered, click on CALCULATE to start the iteration or RESET to reset all initial values.
