THE HRP2 ASSAY:     STANDARD OPERATING PROCEDURES (SOPs)

There are essentially 2 different SOPs available for download as PDF files:
1.) The SOP for the HRP2 Lab Test. This SOP covers the HRP2 drug sensitivity test for culture adapted parasite strains (i.e. for P. falciparum samples that are being kept in continuous culture). It includes washing, diluting the samples with uninfected RBCs, and uses 10% serum in the medium.

2.) The SOP for the HRP2 Field Test. This SOP covers the testing of fresh P. falciparum samples (mainly for field use). The culture procedures are similar to the WHO microtest, with the exception of using 72 hours of culture. The reading, however, is semiautomated and can easily be done in the field with a simple ELISA plate reader. The HRP2 field assay uses a culture procedure that further simplifies the handling and culturing of malaria parasites in the field by omitting centrifugation, washing, the use of serum, and dilution with uninfected red blood cells.
3.) NEW: The SOP for the new ELISA. These draft SOPs cover the new, cheaper in-house ELISA based on 2 commercially available anti-HRP2 monoclonals. Although very convenient, the use of commercial ELISA test kits also made the HRP2 assay relatively expensive. We have therefore developed these SOPs for a simple double-site sandwich ELISA based on commercially available monoclonals from ICLLAB that will cut the price of the ELISA by around 80%.
