Autoimmune mediated renal injury
We are trying to understand mechanisms responsible for glomerulonephritis, a group of severe immune mediated renal diseases that commonly cause kidney failure. Together with Professor Renate Kain, our work concentrates on the origins of autoantibodies to glomerular antigens and the effector mechanisms they engage.
Its ultimate goal is to develop pathway specific treatments for these diseases to replace the non-specific immunosuppressive drugs therapies currently used.
We focus on antibodies to human lysosomal membrane associated protein-2 (LAMP-2) using samples from patients, human and murine in vitro systems and rodent models to analyse: (i) molecular mimicry between LAMP-2 and the bacterial adhesin FimH; (ii) the influence of antibodies to LAMP-2 on macrophages and dendritic cell function, including antigen presentation; and (iii) the pathogenetic effects of anti-LAMP-2 antibodies both in patients and in experimental models.

Contemporary molecular and cellular biology techniques with State of the Art imaging including live cell imaging, confocal microscopy and transmission and immuno-electron microscopy; in vitro and in vivo immunological assays; Flow cytometry RNAi approaches, FACS, and mouse molecular genetics tools (knockout and transgenics).
