Signaling of kinases and Rho GTPases in time  & space: relevance for migration  and proliferation of cancer cells

Traditionally, signaling has been studied mainly by focusing on temporal dynamics. However, mounting evidence in the past decade strongly advocates for the importance of studying the spatial control of cellular signaling. Organelles of the secretory pathway compartmentalize the cellular cytosol and thus it is not surprising to find that a wide range of signaling molecules localize to endomembranes. These membranes may receive signaling inputs and thus be regulated by signaling networks.  Conversely, they may also be the source of signaling pathways. Recent findings on signaling of Cdc42 at the Golgi apparatus and its role in the regulation of cell polarity and directed cell migration will be presented, as well as how this spatial signaling system may be deregulated and involved in cancer.
